You can either fly there from SJU, or take the ferry from Fajardo. The ferries sell out quick though, so get there REALLY early. It is cheap though,2.25 each way. However, the earliest ferry to Culebra is at 9 am, and the latest one coming back is 5 pm. I would definitely suggest spending the night there in Culebra, so you won't have to rush and worry about missing the ferry.
