Gonder is in Ethiopia, and Kassala is on the Eritrean border. To get to Gonder from Sudan, first you have to go to Gedarif (plenty of buses from Khartoum, Medani, Kassala), then change to a local minibus for the border at Gallabat. I have not done this trip, but it is a fairly well-travelled route and shouldn't be too difficult.

As Maykal said it is quite easy but will take 2 days, overnighting in the filthy Ethiopian border village of Metema.
Buses from there to Gonder leave early next morning.
