Depends on where you are staying in Bangkok but generally I would recommend the train (moreorless hourly from the Central station) as bus station is some way out of the centre of the city. Additionally, the bus (eithe direction) can get caught p inthe gridlock that constitutes the city.
Tourist sites are very spread out in Ayuthaya as such there's a lot to see (assuming you aer going just for the day). You may want to hold out for the street market stalls, which open early evening (lot of fun, plenty to chose from).

dear kylian,
there are a few 'best' ways to get to ayuttaya from bangkok. 1. take a boat from the oriental hotel to ayuttaya on the oriental princess with nice lunch on board. 2. take a bus from bangkok , about 1 hour (VIP bus) 3. taking the train, not a bad experience at all and it's about 2 hours. some scenic routes... good restaurant within the touristic area...ummm...got to check that out for you. but there's one restaurant about 10 mins. by car to the other side of the river...run by an influential family with an in house musuem of the ayuttaya heritage....(that got to check the name too, but worth a try!..great seafood !)

Any else to say??
I went by train,it was a pleasent trip you can do the visit of the site in one day going in the early morning and returnig when you finnish sightseeing, we hired a tuk-tuk there and was great, that allows you more time to visit, because the temples are spread on a extended flat area, maybe you can also find some bikes to hire, at Lonelyplanet Thailand there was a very good map of the site, and I'm sure you can get one there.

Thank You guys to this helpful suggestions, I would probably take a train as majority recommended.
Winston, I dont understand the VIP bus which takes an hour travel to Ayuttaya while train takes 2 hrs.? do you mean it is a ''For Hire'"bus that could take you non stop straight to Ayuttaya? the boat from the hotel is quite interesting too with lunch on board, will take a look on it.

Tuk tuk is quite interesting to hire and experienced in Thailand, but could we trust those Tuktuk drivers? I have read many bad comments regarding those Tuktuk drivers here in 'VT Tips Warnings and Dangers'...Well, I do hope that there are some more of them we could trust. Thanks to this tuktuk suggestion, will try!

One bus (VIP) stops inside the park, the cheaper bus stops outside the park. If you go by bus (I did and no problems, North bus terminal) the park bus stop (VIP) has a bicycle rental and internet café plus food. If you arrive and stay inside the park no need to hire tuk-tuks. You will be at the back end of the park, but an easy walk to all attractions. There is an admission fee of 20B approx. for each temple. Food, water, film are in easy reach all over. The only problem I had was with tuk-tuks.

We went one way by boat and return by bus, but this was a real "touristry trip" where as it sounds that the train gets u to where u want to go to spend more time there.
We were fortunate to witness a local wedding with a lot of clour. Tip- Do not eat lettuce, or ice as this is a very easy way to get Salmonela, unfortunately we found out the hard way, even though we new the dangers, never thought about lettuce, and the ice came after too many whiskeys on a rice barge, brain dead..
