Hi Harish,
I'm unsure about buying your ticket in advance or online. Maybe other VT'rs can help you with this.
I have travelled through PP airport twice, and have not noticed any bus ticketing office there. The SR bus does not go past the airport. I have used Mekong Express from SR to PP and they were very good. US$10 and took around 5 hours. I have also used Paramount Angkor Express from PP to Sihanoukville, they also do the PP-SR trip, also a comfortable journey.
Both bus companies leave PP from the riverfront area, and I know that Mekong Express has 2 departures each day, around 7am & 12pm.

You would be very unlucky not to get a seat. Plenty of booking offices in PP along the riverfront, and also around the backpacker area at Boeng Kak Lake. My trip to Sihanoukville included tuk-tuk pick-up from my hotel, I didn't book till the evening before travelling. So you should be OK to book when you arrive in PP. Or, spend a day seeing some of the sights in PP. A very interesting capital city IMO.

I agree with Betska. I know it is a while since I was in Cambodia, but I had no problems at all booking through hotels I was staying in, even at short notice. There are also numerous travel agencies who can assist also. I do not think you will have a problem.

Mekong Express and Sapaco buscompanies are the best. Nice seats, good working A/C. You pay a few USD more for these buses compared to the others. But the other companies are also OK. You can book at one of the agencies on Sisowath Quay (Riverfront) or better ask at the reception in your hotel. If you book 1 day in advance is more then enough. There are several companies who service the line Phnom Penh - Siem Reap . It will take 5-6u to get to Siem Reap.
I traveled a lot on the bus when I visited Cambodja last february.

In Mar 09 the Mekong Express bus ticket from Phnom Penh to Siem Reap was $11. My hotel charged $1 to reserve and pick up the ticket for me. There were four departures from Phnom Penh to Siem Reap each day. I'll e-mail you a picture of the sign. See also my Phnom Penh Transportation tip. The 12:30 PM bus was about 3/4 full. It was a very nice trip. I have it documented under Khett Siem Reab. The ticket even included a nice box lunch and a bottle of water.
