As soon as you arrive in Cusco, you can definitely feel the effects of the high altitude. I felt light-headed the moment we landed at airport. Just take it easy for the next few days. You can properly acclimatize in Cusco for about 2-3 days, but if time is a factor, just proceed directly to lower ground (i.e., Sacred Valley, Urubamba, Ollantaytambo, Aguas Calientes, Machu Picchu) and start enjoying your trip, then you can just go back to Cusco after for sightseeing.
