Hello,
In April I will visit Buenos Aires - Uruguay - The Waterfalls in Foz with my parents. We will arrive in Buenos Aires. After spending a few days in Buenos Aires we will go to colonia and after spending a day in colonia we will take the bus to Montevideo. I really want to go to Cabo Polonia for a couple of days. Is it possible to go from a city close to Cabo Polonia to Foz or should we return to Montevideo ?

When I've been to Cabo Polonio I had a daytrip from Punta del Este, I liked very much, lot of sea lions.
I'm afraid from Cabo Polonio to Foz you should go back to Montevideo then to Assunsion (there is a direct bus) and go to to Ciudad del Este which is close to the border and catarattas. http://www.egakeguay.com

Unless the schedule has expanded, that bus only runs a couple of days a week. You most likely would be better off just going back to Buenos Aires and taking the bus from there.

If you want to go to Foz do Iguazu from Cabo Polonio, I'd do the following:
- take a bus to Chuy (or Chui) in the URU/BRA border and then another to Porto Alegre. There are buses going Monetvideo - Porto Alegre, stopping in Punta del Este if you prefer.
- From Porto Alegre, take another bus to Foz do Iguazu.

Perhaps it's a longer way, but faster connections as there might be better bus frequencies between those cities rather than going from Montevideo or Bs.As.
From Cabo Polonio to Porto Alegre is only 550 kms aprox.
