The wineries are located between Santiago and Viña in the Casablanca Valley. The only one I have actually visited was the House of Morandé and I really went to the restaurant only. It is possible to visit the wineries in the area though however this would complicate things with regard to your transportation between Santiago and Viña as the buses I recommended do not stop here.
Once you are in Viña I suggest that you spend at least a day walking around different parts of Valparaiso. Cerro Concepcion and Cerro Artilleria are two of my favourites. Take the asensores up the hills and walk around for a bit.
In Viña itself you could go to the flower clock and perhaps walk around the hill above it where there are some historical buildings and castles. Unfortunately they aren´t open for public viewing. You should also go to the Muelle Vergara (which is closed) and walk from there along the boardwalk and look at the craft markets there.
I suggest you take a bus or taxi to Higuerillas (on the way to ConCon) and have a seafood lunch there.
