I plan to go to Siem Reap from Phnom Penh by bus. I'll go there in early Oct.
How early do I have to buy the ticket in advance, or I can just buy it when I get there (for next day trip)? According to my readings, Mekong Express seems to be the best and the trop takes about 5.5 hours. Do they sell round trip ticket?

better buy your ticket 1 day prior departure, some how the office of Mekong Express is conveniently located at the river bank.

Vanakam, I suggest you buy the coach ticket on your first day in PP itself.

Depending on where you stay, your hotel/guesthouse tour desk may be able to assist you with this, you just need to pay them the service charge (probably 1 or 2 US$).

That gets 1 concern off your mind & you can enjoy the remainder of your time in PP without further concerns.

like other posters said, earlier is better. Especially Mekong Express ticket is very well selling.
I could not get Mekong Express ticket when I was in Phnom Penh at 2006. At that time I need a same day departure for 7 persons. No wonder yeah.
However, not far from Mekong Express, only a couple of shop away, there was another bus operator ticket counter. Unfortunately today I forget its name, but that counter was near a restaurant in the corner of the block. There I could buy 7 tickets for a same day departures.
The condition of the bus is OK for me. There were several caucasian tourists, but the majority of the passengers were Cambodian.
Along the journey we can saw video of Cambodian video klip and Cambodian movie. So, that 6 hours journey was not a boring at all.

I would recommend Mekong Express as the best bus company. They are reliable, they pick you up on time unlike other companies. They give you a commentary on sights of interest along the way. You get sandwiches and water. The staff are really friendly and helpful. We were waved off by the whole office once. We didn't book direct but used a travel agent. We booked tickets for the next day without a problem. There were two of us.
