1. You should post these questions in the appropriate Travel Forums (Barcelona and Rome, or even Spain and Italy), because most VTers don't read the Technical Forum, as it is dedicated to questions about using the VT website, not travel questions.
2. The rail website for Spain is www.renfe.es . Look for "Seleccione su idoma" on the lower left for English. Note that this is not the most user-friendly website, but you will be able to answer the Barcelona to Madrid question there.
3. The rail website for Italy is www.trenitalia.com/en/index.html (this is in English). So far as I know, there is no longer a direct train from Rome to Barcelona, but the trip will require 2 or 3 changes. Because of this (and the duration of the journey), flying is normally the preferred way to go for such a long distance.
4. When you use websites to price things, choose a date in the next week or so that is the same day of the week as the date you plan to go in October. Rail websites in particular are not accurate more than 60 days out, and often won't show you any solutions at all. Although schedules can change over the year, they don't change all that much, nor do the fares.
