The nearest brazilian infrastructure to Manu and Puerto Maldonado is a roadway.
This road connects Bolivia (very near to Peru) with Brazil and serves the brazilian city Assis Brasil in Acrean brazilian state. Assis Brasil is a triple border Brazil/Bolivia/Peru. From Assis Brasil you may go to Iņapari/Peru and get a taxi to Pueto Maldonado. The brazilian part of the road is paved. The other brazilian crossing to Peru is by BR-364 near to Rodrigues Alves city. I believe between Assis Brasil and Rodrigues Alves the only transportation is boat. The jungle reigns there.

Are you going to drive or fly? I don't think driving would be a short trip. I think you can fly out of Cusco to Puerto Maldonaldo, spend 2 night at Cortes Maltes and return satisfied. I'd also reccommend you check out Cesar at Lamanita Travel.com.
