There are 5 rail solutions a day from Segovia to Salamanca, according to the German rail website www.bahn.de (I use this because the Renfe (Spain) website has become more useless every time they 'update' it).
Each solution has a change of train in Villalba de Guadarrama...essentially, you take a local train from Segovia towards Madrid, and you get off early, and catch a faster train leaving from Madrid out to Salamanca.
The various rail solution are 3 to 4.5 hours...you could drive it in 2.25 hours, which makes you wonder if a direct bus would be faster...but I don't know of one...maybe someone else will chime in...
