Probably not although the tourist office there might be able to help. Many places want min. 3 night stay. I stayed at Hotel Ca d'Andrean http://www.cadandrean.it/ - very nice, very reasonable, very clean in Manarola. I was able to book 2 night there. If you can't find anything go to the tourist office - they can help - just make sure you see the place before paying. I have heard stories of wonderful places and dumps when people did this.

Cinque Terre are located in Liguria, not Tuscany...

Rita, I was wondering if there were some other Cinque Terre I hadn't heard of!
About accommodation - the good thing is that you've not hit the busy season yet. I think your chances of finding something, even on the day, are very good. Especially as, in addition to guesthouses, local people rent out rooms. I stayed at L'Eremo Sul Mare, a b&b just outside Vernazza and would really recommend it. Great hosts, rooms, location.
