by train to Naples and then bus or taxi to Positano or, once in Naples, take circumvesuviana (a small railway connecting cities near Naples) line 1 to Sorrento and it would be easier to find bus/taxi to Positano
As far I remember there is also a direct flight from Venice to Naples...

Train to Naples:

http://www.trenitalia.com/en/index.html

Circumvesuvia (a private line) train to Sorrento (from the same station, though you'll have to walk downnstair I believe):

http://www.vesuviana.it/

About an hour on the train will bring you to Sorrento. Buses to Positano go from directly outside the station. If you walk straight across the (small) road, down the steps across the road again and look to your right you'll see the timetable posted bu the wall (there will be several buses hanging around the area).

The SITA bus timetable is online at

http://www.sitabus.it/wps/portal/OrariCampania

but down at the moment. When it's up you will need the 'Quadro 15' Amalfi-Positano-Sorrento' timetable.

If you are desperate to know the timetable now, this site has a listing (it also has awful music, sorry!):

Time your journey carefully, because missing the last bus will mean an expensive taxi ride (or a night in Sorrento).

welcome to VT. Without knowing any more than your question, have you considered renting a car? A relatively easy drive, mostly toll free autostrada, scenic for part of the trip, and greater convenience and less reliability on public busses while you are there.

There is a ferry from Naples to Positano which gives you the most glorious sights to see including the Isle of Capri and Vesuvius. It runs from the Port, just ask anyone to direct you to the ticket office. We went on a ferry at about noon and had about three in Positano before returning on the ferry to Naples. It was absolutely beautiful.

That should read "about three hours" in Positano.
