Metro del Mare is suspended at the moment but service starts on 31/3 - however, there are only 2 runs a day from Naples to Positano, at 14:15 and 19:15, see www.metrodelmare.com and click on timetables then low season (MM2).
Otherwise, you can take the Circumvesuviana train from below Napoli Centrale station to Sorrento and SITA bus from there to Positano (there used to be the odd SITA bus all the way to Positano from Naples' Piazza Municipio but I couldn't find it on current timetables http://www.sitabus.it/wps/portal/OrariCampania).
If you are arriving at Naples airport, there are buses direct to Sorrento, as well as the city shuttle that stops at Napoli Centrale.

okay, in rough weather, the ferry has to bypass positano which has an unprotected pier and go directly to amalfi. If that happens, the SITA bus terminal there is right across from the harbor and you can get a bus back to positano. however, having done this, not recommended.
i have rented a car on the amalfi coast and really think that is the best way to get around. the bus schedules are great, but some people have found them less than accurate. with the car, you can drive easily to places like ravello which are breathtaking.
if the car is not for you, then the advices above on the Circumvesuviano train and the SITA bus are about the only other option, besides an expensive private driver.

The bus from Sorrento to Positano (Amalfi actually) is a fascinating experience in itself......so much so that I have made specific travelogues about it in my Amalfi page! It takes around an hour; buses go from directly outside the Sorrento Circumvesuviana station.

Circumvesuviana train timetable here: http://www.vesuviana.it/

Bus timetable here: http://www.sitabus.it/wps/portal/OrariCampania Scroll down to Quadro 15 for Amalfi-Positano-Sorrento details.
