How much will it cost to take a taxi from the Tunis airport to the Sheraton Hotel and Towers? Has anyone ever stayed at this hotel? Is it centrally located? Also, how much should it cost to take a taxi from the Sheraton Hotel to the souks at the Medina?

Usually any transfer from the airport will cost a tourist 10TND. Try to bargain it down to 5TND as the Hotel is closer to the Airport than Hotels in downtown Tunis. It's not downtown, but just north of it and the Belvedere park.
Sheraton hotel to the Medina will cost roughly 3 TND... you never know with the traffic in downtown.
