You may want to consider Milan as a hub. So you could take the overnight train from Barcelona to Milan http://www.renfe.es/horarios/index.html
of fly from Barcelona to Milan www.vueling.com.
Milan is close to Florence so you could visit Florence when you get to Milan. Have you considered visiting Lake Como? Short hop from Milan, very enjoyable - (shameless plug) - I have some photos and restaurant tips for Varenna, off Lake Como on my pages.
The Nice/Monaco stop would be when you're in Cannes - I would also suggest stopping at Eze, which is on the way to Monaco. There are trains between Cannes, Nice, and Monaco, you just need to double check there are no rail strikes when you're visiting.
I know you have a rail ticket, but not knowing how long you have, flying could save you some time. If time is not an issue, do keep to the train, but note that many times larger cities close to the places you want to visit will have better connections, which is the case with Milan.
