1) By bus from BKK, you can catch the bus at Aekamai (Eastern Bus Terminal), buy a Bangkok-Ban Pae ticket (around 100 Baht) . It will take you 3 hours to get to Ban Pae where you can take a boat to Samet island, the pier is just a cross the street from bus terminal.
2) I think from March to June are the best.
