By rented car is the best.
Beware this road is the baddest, but also very beautiful, in whole Cuba, so take your time Its possible to rent a car from $45 USD a day Anja

Thank you Anja. I use to travel by Jeep when I was a child, indeed is a bad terrain to travel on, but I though that maybe things had improved or had a bus route. Thank you again.

There's no bus, of course you can hire a jeep or a car, but the cheaper and more adventurous way is going in a 'camioneta'. I did it from Baracoa to Moa. It's NOT comfortable (no so far either), but it's an amazing experience meeting all these Cubans piled up in such old vans. And it's even illegal, but it will cost you 5 or 10 dollars at most. Try to leave as early as possible, 6 a.m. or so. There's bus station in Moa where the camionetas leave.
