Do you means private audience? That is very difficult for obvious reasons. Through your local church would be the way to go but it would need to be for a very special reason.

However the Pope does give a general audience most Wednesdays - tickets are required but they are free of charge. Your otel in Rome shoud be able to get you a ticket if you let them know well in advance. There is also the Angelus blessing at noon most Sundays, for which no tickets are required. Both obviously depend on the Pope being in good health and in Rome at the time.
