May I know the time of the earliest bus departing to Melaka from puduraya in KL and what time the last bus from Melaka to KL.

Earliest bus to Melaka is at 8.00am abnd last bus to Kl is at 10pm if I'm not mistaken. You may check Transnasional's website for details: http://www.nadi.com.my/transportation_routes_express.asp -m-

Is it necessary to book in advance? Or can I just show up at the station?

To be safe its better to book in advance if you are travelling during the peak season (weekends, school & public holidays).
