I'm not sure if 191 is the best way, especially if you want to make the trip as quickly as possible. Whatever way you go, it's a long drive for 1 day. I checked mapquest.com and put in Moab to Jackson. Mapquest suggests taking 191 to Price, then the 6 to Interstate 15 which takes you north through Salt Lake City and into Idaho. Then you can cut across into Wyoming. I have also taken Rt 89 through Logan and around Bear Lake into Idaho and then into Wyoming...
The route that mapquest recommends is 535 miles and estimated at 9 hrs drive time. But, if you hit Salt Lake City around rush hour - you can add on some time for sure...
I just checked mapquest again between Moab and Vernal and Vernal to Jackson which would be 191 all the way. It's actually not too much different... It adds on an estimated 55 minutes but is 30 miles shorter than taking I 15 into Idaho.
Out of curiosity, I also checked mapquest for the route I usually drove between SLC and Jackson which is 89 through Logan (north of SLC) and Bear Lake. Going that route starting from Moab would also be 9 hrs and is 5 miles shorter than going 191 all the way.
So, there isn't too much difference in these 3 possibilities...

That's a little hard to answer because I don't know what you mean by best. If you would like the most straight-forward, fastest then you should probably go to SLC then north on 1-15 to Idaho Falls then east on 26 to Jackson.

If you want the most scenic then head east from Brigham City (north of SLC on I-15) and over on 89 to Logan/ Bear lake/Jackson.

If you want the one that is the least traveled then stay on 191 the whole way. It is 2-way traffic, but good roads. Very few cars on the road between Helper and Duchesne. You could stop in Vernal to look at the Dinosaur Museum and Monument. You could stop at Flaming Gorge for some nice fishing, picnicing, and the road from Rock Springs to Pinedale has some great views of the Wind River mts. I'd probably do it just for the novelty, but that's because I've gone all the other ways. I've been on all the parts of that road, just not all together.
