I have been in to Naples on various cruise ships and I'm fairly sure you can travel at least part of the way to Positano by ferry or hydrofoil. We always visit Capri and I know there is a connection between Capri and Positano in the summer months (not sure when they start). Make sure you visit Capri - it's beautiful. Start off at the waterfront where all the ferries come in and then go up to the main town, by either bus, on the cable car or by stretch taxi. The view is spectacular, the shops are great (from cheap handmade sandles to designer clothes) and there are lot's of little alleyways to explore and get lost in! From there go by bus or taxi to Ana Capri, down the far side of the island. There are some great bars and eateries right on the pebble beach. I know one of the ferry companies is Lineajet and I'm sure you could check out their timetables through a Google search.

By May you should be able to take a passenger ferry from Naples Beverello port to Positano. It's faster and more scenic than the bus. There isn't train service. Bring sunscreen.
