In addition to the urban buses connecting the airport to downtown Valladolid mentioned by Revulgo, there are also buses going all the way to Madrid (2hrs) and Leon. Tickets must be purchased in advanced on www.alsa.es
I do not find Valladolid particularly fascinating. To me, it is rather an ugly sprawling modern city, even though the area around the main square (Plaza Mayor) and the cathedral is kind of cute. Still, the city could not be better located, almost equidistant from some of the most interesting historic cities in central Spain: Leon, Salamanca, Avila, Segovia and Burgos. You could easily base yourself in Valladolid and take day trips to some of these cities. Salamanca and Segovia are absolutely amazing.
Vitoria is quite a long way from Valladolid, about 3hrs by car. Not sure about public transport.
