Surely by train, but you have to change in Florence, so Siena -> Florence / Florence -> Venice.

The train is definitely the easiest way to go. If am correct the location of your hotel is east of the Piazza di San Marco so use the vaporetto to get from the train station to the S. Zaccheria stop if you have luggage.

I would not suggest attempting to walk through the city to your hotel dragging luggage around because although the numerous bridges and narrow streets are interesting when you are sight seeing, they become a serious problem with baggage. Although if you feel really extravagant you can hire a taxi to take you to your hotel but that will cost you...
