To reach San Gimignano from Firenze by bus you have to take it at the Autobus Station in Florence in Via Santa Caterina da Siena in front of Santa Maria Novella Train Station and go to Poggibonsi. In Poggibonsi you'll take another bus to San Gimignano.

Here timetable and some information about San Gimignano, a lovely town in my opinion.

SITA bus has a day trip to Siena and San Gimignano which is very good. This includes an English speaking guide. Check out their web site: www.sitabus.it. Click on Turismo then Escursioni. You then get the option to view the page in English.
Their sales office is located near where you catch the regular busses, and the sight seeing busses also leave from that terminal.
