I went 2 time to Ayuthayya and both of the times are by bus from MoChit Bus Terminal. So I guess Bus is better. Once reachaed Ayuthayya we hired a over there. Few personal mini tuk-tuk like will be waiting there. They will hold all the major tourist places post card pictures. Maybe around 10 places if I am not mistaken. So to see all of them will consume around 5 to 6 hours. So we hired him to take us to all the places. Regardless how many people we are for those 5 to 6 hours we payed 500/-THB. You can bargain. One of the driver mobile no. and name is Ti - 051897239

HI! Mate:

The tuk-tuk's fare is regulated but you can of course bargain, if you are touring longer or if you have more than 1 traveler.

Maybe you'll like to check my VT Ayutthaya page for more details?

3 of us rode around all day and, were picked back up the following day and transported to the train station, along with our baggage.
We paid $20.00 for the day/night for use of the tuk tuk and their services and, tipped generously as well.
I'd say around $20.00/day is a good base price.
