It all depends on your budget. Flying is an option Siem Reap - Bkk - Trat but this is going to cost a minimum of 8,000 baht/person when you include departure tax and getting to your hotel on Koh Chang.
The other way that is hassle free is to arrange aprivate transfer by car ( if there are only 2-3 people) or minivan if there are more. Total cost from Siem Reap to any resort on Koh Chang is 8,000 baht for a car, 9,500 baht for a minivan. It would take the best part of a day - leave Soem Reap after breakfast - say around 8-9am and be on the 6pm or 7pm ferry from the mainland to KC. You probably could do it cheaper but this is with transfers completed by honest, reliable tour agencies who work with large hotels on KC & in Cambodia - not with companies who do thing as cheaply as possible.
