Both are absolutely worth a visit.
Two hours is an absolute min. for Corinthos. But if in addition to classical Greece, you are also interested in early Christian history, you will spend much more time here.
Three hours is a min. for Mycenae.
Bus timetables and prices here: http://www.travelpage.gr/transportation/trans_buses7.php.
How to get to Corinthos and Mycenae, please also see my post here: http://forum.virtualtourist.com/1/discussion-223235-1-5-Travel-0-461-Athens-discussion.html?wosid=KoWZ7Pv18ZZwt09Owd5DKM
GKE's suggestion to make this a two day trip is very good:
leave Athens in early the morning, visit Corinthos, continue to Nafplio.
Visit Nafplio in afternoon, stay overnight in Nafplio.
Next morning, visit Mycenae and return to Athens.
Note this does not allow time for a visit to Epidavros, which is very much worthwile as well. Add one more night in Nafplio to visit Epidavros.
