my advice is go by bus.
they are cheap, comfortable and safe. travelling by private car is not recommended in brazil in my opinion.

If this is your first time in Brazil, don't drive, especially such a long distance. Take the comfortable bus between cities,as recommended, or fly. Then you can take day trips from both cities.
