My husband and I would like to visit Athens, Crete, Santorini and Levros in October for 14 days (this doesn't include round trip flight time from the U.S.), but Levros seems a bit out of the way. Would it be practical to try to set this up and/or do you think Levros is worth including or unique in some way. We will be doing car rental except in Athens. Thanks you for your advice!

I have been to Lesbos however this was part of a N.E. island hop, I would never try to include it with Santorini or Crete. There is no point in including it with your other destinations. Athens, Santorini, Crete will be more than enough for 14 days.
Lesbos is very worthy of a visit, however it is not really unique, just that it would be more practical/easier to go something like Athens-Lesbos-Chios-Samos-Athens.
