We, a group of 5 people are planning to travel to MP in mid Oct. I have afew questions:
1- Will the weather be fine from Oct 15- Oct 23?
2- To complete the trail, do we have to carry all the supplies (Tent, Sleeping Bag, Food) or are there any rest-stops on the 4 day trail?
3- Do we need to get a license for the trail? Is there a quota? Where do we apply to get access to the trail?
4- Once back, how can we go to Lake Titicaca? Are there flights or buses? Cost?
Any other advise will be appreciated.

Having just come back from there a few weeks ago I can tell you that:
Mid October you should just be ok with the weather. Rainy season starts end Oct/Beginning Nov.
You can hire porters to carry some of your gear, however there are weight limits they are allowed to carry.
You now need a permit, pre booked, to do the trail. You can't just turn up and go. You can probably find someone who can arrange this on the web. Also, make sure that the details on the permit EXACTLY match that on your passport. One guy was turned away because of one number being out on his passport no.
There are buses that go from Cuszco to Puno on Lake Titicaca, the journey is around 6 hours. I don't have details but you can probably find something on the web or go to one of the many tour offices in Cuzco when you get there.

The only other thing I can add to mad4travels advice (which is right on the money) is that you can fly from Cuzco to Juliaca (which is very close to Puno). The flight is 50 minutes long, but will be much more expensive than the bus.

I booked my inca trail a month before I arrived in peru. I booked mine through Axel Tours (www.axeltours.com).
All the tour companies provide porters to carry tents and food but you may be expected to carry your own sleeping bag and mattress.
I got the bus from Cusco to lake to Puno which is on the shores of Lake Titikaka. The trip took about 6 hours and cost around 7 USD.

Because no one mentioned it yet - you don't just need a permit, you have to hire a guide for the trail. You cannot do it on your own.

Depending on which company you choose porters may or may not carry your bags. Generally they will carry all supplies and camping gear. The company I hiked with (BigFoot Tours) did that, and you carried your personal supplies. If you wanted you own bag carried you payed a premium.
As far as getting to Lake Titicaca...If you are doing the 4 day hike, by the time you tour MP on the last day and catch the train back to Cusco it will be late after noon early evening. There are overnight buses that will take you to Puno or Copapcabana (which I recommend over Puno, check my pages). What I did was prepurchase these tickets before i hiked the trail to guarantee my spot. Alternatively you can take you chances or take one of the many buses the next day. The route is very scenic, so if you can afford the time, a day time bus trip may be worth it.
