This is the whole question- not part like I previously posted! We are choosing between the Domus Henrici, the Ibis Praha Old Town and Hotel 16 for two nights in Prague. Other then proximity to the sites, we're looking for ambience of the immediate surroundings of the hotel. Any ideas? Thank you!
My opinion only (solo middle-aged female, visited Prague twice so far): Hotel 16 is too far out for enjoyable wandering and exploring of Prague's historical centre. It's a pleasant enough area but I stayed near Metro IP Pavlova the first time I visited Prague and realised I would have much preferred to be nearer the old centre. Domus Henrici is also quite some way from the Old Town (you'll definitely need a tram or taxi), although nearer the castle. I haven't been as far along that road as the hotel's location, so do not know what the actual area is like, but I would not choose it for that reason. The Ibis is within easy walking distance of the sights/sites of that location and trams to Mala Strana, the castle etc....and you have a vast choice of restaurants/cafes/bars/shops etc nearby. So, for only 2 days' exploration and all other things being more or less equal there would be no contest. I would choose the Ibis.

Stayed at Ibis last year. No complaints.

Hotel 16 is in a rather dull area, busy streets with high air pollution not far away. Ibis Praha Old Town is on the one of main shopping/tourist streets. So you can join the crowd as soon as you step out of the door. Domus Henrici is somewhat away from the center, but you can take relaxed walks to the Castle, Strahov monastery, Petrin hill, Mala Strana. I would say this one has best ambience.

Not too sure about what you mean by "ambience". Top of my list of criteria is "safety". Never felt unsafe in Old Town. By the way, if you are interested, have a review of Ibis here: "Central Location" - Ibis Praha Old Town Tip by pedroswift with a couple of nearby restaurants as well on the Prague page.
