Let me modify the question a bit, How much do the buses charge from Port Elizabeth to Grahamstown and at what time do they operate? Is there a train service between the two towns and if so how much does it cost?

There is no train service between the two cities. As Grahamstown is a university town there are a few operators who offer transfers. Those are not that cheap, so all depends a bit on your budget.
