You can take a bus to Plymouth Center from Boston (Plymouth & Boston Bus Lines), but then you'd have to take a taxi from there. It's really much easier to rent a car for the day.

I went to their website and they have a comprehensive list of how to get there from Boston:

    Public Transportation from Boston:
Public transportation in Plymouth is currently under redesign. We at Plimoth Plantation strongly urge our guests to rent a car for their visit to Plimoth Plantation and Mayflower II. If a one day car rental is not an option then listed below are the most up-to-date resources available.

From Boston's South Station, take the Commuter Rail (1-800-392-6100) train to the North Plymouth station. This is an open platform station. From the North Plymouth station you will need to arrange for a taxi service or use the local GATRA bus service (GATRA is designed for residents of the town and may prove problematic for tourists). Taxi ride from the North Plimoth station to Plimoth Plantation is about 15 minutes or Mayflower II about 8 minutes.

    Sea Breeze Taxi (508) 888-0774

South Shore Taxi (508) 406-8908 or (781) 535-4405 are available 24 hours a day. Also offering van service to and from TF Green and Logan airports.

The local GATRA Bus line (508- 746-0378) runs between Plymouth center and our the museum.

Pre-arranged shuttle service available by Access Express (508) 746-5715.

You may also travel by the Plymouth-Brockton Bus line (508-746-0378) from Boston to Plymouth which stops at exit 5. You will need to use the GATRA bus service or one of the listed taxi services.
